Principle of the Assay: The Human EDIL3 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human EDIL3 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human EDIL3 coated on a 96-well plate. Standards and samples are pipetted into the wells and EDIL3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human EDIL3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of EDIL3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background: EDIL3, also known as DEL1, is a glycoprotein that is secreted by endothelial cells and can associate with the endothelial cell surface and the extracellular matrix. It is regulated by hypoxia or vascular injury and has been implicated in vascular remodeling during angiogenesis. The EDIL3 gene is mapped to 5q14. Human endothelial cells bind to Del1 together with the alpha-V (ITGAV)/beta-3 (ITGB3) integrin receptor. It is confirmed that adhesion of endothelial cells to Del1 is integrin mediated and that Del1 binds ITGAV/ITGB3. Functional analysis showed that both Del1 and vitronectin (VTN) stimulate endothelial cell migration. Immunoprecipitation and immunoblot analyses revealed that adhesion of endothelial cells to Del1 induces tyrosine phosphorylation of PTK2.
