Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human DKK3 antibody. DKK3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human DKK3 Antibody is added and binds to DKK3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated DKK3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human DKK3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

This gene encodes a protein that is a member of the dickkopf family. The secreted protein contains two cysteine rich regions and is involved in embryonic development through its interactions with the Wnt signaling pathway. The expression of this gene is decreased in a variety of cancer cell lines and it may function as a tumor suppressor gene. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease ().
