Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for ACO1 elisa kit
Description: This Human Cytoplasmic aconitate hydratase (ACO1) ELISA Kit employs a two-site sandwich ELISA to quantitate ACO1 in samples. An antibody specific for ACO1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ACO1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ACO1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACO1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Aconitase 1 is a cytosolic protein which binds to iron-responsive elements (IREs). IREs are stem-loop structures found in the 5' UTR of ferritin mRNA, and in the 3' UTR of transferrin receptor mRNA. The iron-induced binding to the IRE results in repression of translation of ferritin mRNA, and inhibition of degradation of the otherwise rapidly degrading transferrin receptor mRNA. The cytoplasmic IREBP interacts with the IREs of these mRNAs. The iron status of the cell determines the ability of IREBP to bind to an IRE through reversible oxidation-reduction of sulfhydryl groups that are critical for the high-affinity RNA-protein interaction. Thus, IREBP plays a central role in cellular iron homeostasis by regulating ferritin mRNA translation and TFRC mRNA stability

NCBI Description

The protein encoded by this gene is a bifunctional, cytosolic protein that functions as an essential enzyme in the TCA cycle and interacts with mRNA to control the levels of iron inside cells. When cellular iron levels are high, this protein binds to a 4Fe-4S cluster and functions as an aconitase. Aconitases are iron-sulfur proteins that function to catalyze the conversion of citrate to isocitrate. When cellular iron levels are low, the protein binds to iron-responsive elements (IREs), which are stem-loop structures found in the 5' UTR of ferritin mRNA, and in the 3' UTR of transferrin receptor mRNA. When the protein binds to IRE, it results in repression of translation of ferritin mRNA, and inhibition of degradation of the otherwise rapidly degraded transferrin receptor mRNA. The encoded protein has been identified as a moonlighting protein based on its ability to perform mechanistically distinct functions. Alternative splicing results in multiple transcript variants [provided by RefSeq, Jan 2014]

Uniprot Description

IREB1: Iron sensor. Binds a 4Fe-4S cluster and functions as aconitase when cellular iron levels are high. Functions as mRNA binding protein that regulates uptake, sequestration and utilization of iron when cellular iron levels are low. Binds to iron-responsive elements (IRES) in target mRNA species when iron levels are low. Binding of a 4Fe-4S cluster precludes RNA binding. Belongs to the aconitase/IPM isomerase family.

Product Notes

The Human ACO1 aco1 (Catalog #AAA9714545) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714545 ELISA Kit recognizes Human ACO1. It is sometimes possible for the material contained within the vial of "Cytoplasmic Aconitate Hydratase (ACO1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
