Principle of the Assay: SPRR1A ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SPRR1A antibody and an SPRR1A-HRP conjugate. The assay sample and buffer are incubated together with SPRR1A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SPRR1A concentration since SPRR1A from samples and SPRR1A-HRP conjugate compete for the anti-SPRR1A antibody binding site. Since the number of sites is limited, as more sites are occupied by SPRR1A from the sample, fewer sites are left to bind SPRR1A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SPRR1A concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
Uniprot Description
SPRR1A: Cross-linked envelope protein of keratinocytes. It is a keratinocyte protein that first appears in the cell cytosol, but ultimately becomes cross-linked to membrane proteins by transglutaminase. All that results in the formation of an insoluble envelope beneath the plasma membrane. Belongs to the cornifin (SPRR) family.
Protein type: Adaptor/scaffold
Chromosomal Location of Human Ortholog: 1q21-q22
Cellular Component: cytoplasm; cornified envelope
Molecular Function: protein binding, bridging; protein binding; structural molecule activity
Biological Process: keratinocyte differentiation; epidermis development; keratinization; peptide cross-linking

Product Notes
The Human SPRR1A sprr1a (Catalog #AAA7244292) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7244292 ELISA Kit recognizes Human SPRR1A. It is sometimes possible for the material contained within the vial of "Cornifin A (SPRR1A), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
