Intended Uses: This C1QA ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human C1QA. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: C1QA ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-C1QA antibody and an C1QA-HRP conjugate. The assay sample and buffer are incubated together with C1QA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C1QA concentration since C1QA from samples and C1QA-HRP conjugate compete for the anti-C1QA antibody binding site. Since the number of sites is limited, as more sites are occupied by C1QA from the sample, fewer sites are left to bind C1QA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C1QA concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a major constituent of the human complement subcomponent C1q. C1q associates with C1r and C1s in order to yield the first component of the serum complement system. Deficiency of C1q has been associated with lupus erythematosus and glomerulonephritis. C1q is composed of 18 polypeptide chains: six A-chains, six B-chains, and six C-chains. Each chain contains a collagen-like region located near the N terminus and a C-terminal globular region. The A-, B-, and C-chains are arranged in the order A-C-B on chromosome 1. This gene encodes the A-chain polypeptide of human complement subcomponent C1q. [provided by RefSeq, Jul 2008]

C1QA iso2: C1q associates with the proenzymes C1r and C1s to yield C1, the first component of the serum complement system. The collagen-like regions of C1q interact with the Ca(2 )-dependent C1r(2)C1s(2) proenzyme complex, and efficient activation of C1 takes place on interaction of the globular heads of C1q with the Fc regions of IgG or IgM antibody present in immune complexes.
