Intended Uses: This CAT ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CAT. This ELISA kit for research use only!

Principle of the Assay: CAT ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CAT antibody and an CAT-HRP conjugate. The assay sample and buffer are incubated together with CAT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CAT concentration since CAT from samples and CAT-HRP conjugate compete for the anti-CAT antibody binding site. Since the number of sites is limited, as more sites are occupied by CAT from the sample, fewer sites are left to bind CAT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CAT concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes carnitine acetyltransferase (CRAT), which is a key enzyme in the metabolic pathway in mitochondria, peroxisomes and endoplasmic reticulum. CRAT catalyzes the reversible transfer of acyl groups from an acyl-CoA thioester to carnitine and regulates the ratio of acylCoA/CoA in the subcellular compartments. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Apr 2012]

CRAT: Carnitine acetylase is specific for short chain fatty acids. Carnitine acetylase seems to affect the flux through the pyruvate dehydrogenase complex. It may be involved as well in the transport of acetyl-CoA into mitochondria. Belongs to the carnitine/choline acetyltransferase family. 3 isoforms of the human protein are produced by alternative splicing.
