Intended Uses: This sandwich kit is for the accurate quantitative detection of human Carbonyl reductase [NADPH] 1 (also known as CBR1/CBR/CRN) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human CBR1/CBR/CRN antibody. CBR1/CBR/CRN present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human CBR1/CBR/CRN Antibody is added and binds to CBR1/CBR/CRN in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CBR1/CBR/CRN antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human CBR1/CBR/CRN. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

CBR1: NADPH-dependent reductase with broad substrate specificity. Catalyzes the reduction of a wide variety of carbonyl compounds including quinones, prostaglandins, menadione, plus various xenobiotics. Catalyzes the reduction of the antitumor anthracyclines doxorubicin and daunorubicin to the cardiotoxic compounds doxorubicinol and daunorubicinol. Can convert prostaglandin E2 to prostaglandin F2-alpha. Can bind glutathione, which explains its higher affinity for glutathione-conjugated substrates. Catalyzes the reduction of S-nitrosoglutathione. Belongs to the short-chain dehydrogenases/reductases (SDR) family.
