Intended Uses: This OPG ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of RatOPG. This ELISA kit for research use only!

Principle of the Assay: OPG ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-OPG antibody and an OPG-HRP conjugate. The assay sample and buffer are incubated together with OPG-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the OPG concentration since OPG from samples and OPG-HRP conjugate compete for the anti-OPG antibody binding site. Since the number of sites is limited, as more sites are occupied by OPG from the sample, fewer sites are left to bind OPG-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The OPG concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a membrane calcium channel subunit that is activated by the calcium sensor STIM1 when calcium stores are depleted. This type of channel is the primary way for calcium influx into T-cells. Defects in this gene are a cause of immune dysfunction with T-cell inactivation due to calcium entry defect type 1 (IDTICED1). CRAC channels are the main pathway for Ca(2+) influx in T-cells and promote the immune response to pathogens by activating the transcription factor NFAT. Interacts with STIM1 and STIM2. Interacts with EFCAB4B/CRACR2A; the interaction is direct and takes place in absence of Ca(2+). Forms a complex with EFCAB4B/CRACR2A and STIM1 at low concentration of Ca(2+), the complex dissociates at elevated Ca(2+) concentrations. Belongs to the Orai family.
