Intended Uses: This CCL7/MCP3/SCYA6/SCYA7 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CCL7/MCP3/SCYA6/SCYA7. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CCL7/MCP3/SCYA6/SCYA7 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CCL7/MCP3/SCYA6/SCYA7 antibody and an CCL7/MCP3/SCYA6/SCYA7-HRP conjugate. The assay sample and buffer are incubated together with CCL7/MCP3/SCYA6/SCYA7-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CCL7/MCP3/SCYA6/SCYA7 concentration since CCL7/MCP3/SCYA6/SCYA7 from samples and CCL7/MCP3/SCYA6/SCYA7-HRP conjugate compete for the anti-CCL7/MCP3/SCYA6/SCYA7 antibody binding site. Since the number of sites is limited, as more sites are occupied by CCL7/MCP3/SCYA6/SCYA7 from the sample, fewer sites are left to bind CCL7/MCP3/SCYA6/SCYA7-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CCL7/MCP3/SCYA6/SCYA7 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes monocyte chemotactic protein 3, a secreted chemokine which attracts macrophages during inflammation and metastasis. It is a member of the C-C subfamily of chemokines which are characterized by having two adjacent cysteine residues. The protein is an in vivo substrate of matrix metalloproteinase 2, an enzyme which degrades components of the extracellular matrix. This gene is part of a cluster of C-C chemokine family members on chromosome 17q. [provided by RefSeq, Jul 2008]
