Principle of the assay: BACE2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BACE2 antibody and an BACE2-HRP conjugate. The assay sample and buffer are incubated together with BACE2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BACE2 concentration since BACE2 from samples and BACE2-HRP conjugate compete for the anti-BACE2 antibody binding site. Since the number of sites is limited, as more sites are occupied by BACE2 from the sample, fewer sites are left to bind BACE2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BACE2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes an integral membrane glycoprotein that functions as an aspartic protease. The encoded protein cleaves amyloid precursor protein into amyloid beta peptide, which is a critical step in the etiology of Alzheimer's disease and Down syndrome. The protein precursor is further processed into an active mature peptide. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2013]

BACE2: Responsible for the proteolytic processing of the amyloid precursor protein (APP). Cleaves APP, between residues 690 and 691, leading to the generation and extracellular release of beta-cleaved soluble APP, and a corresponding cell-associated C- terminal fragment which is later released by gamma-secretase. It has also been shown that it can cleave APP between residues 671 and 672. Belongs to the peptidase A1 family. 5 isoforms of the human protein are produced by alternative splicing.
