Intended Uses: This BCAT2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse BCAT2. This ELISA kit for research use only!

Principle of the Assay: BCAT2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-BCAT2 antibody and an BCAT2-HRP conjugate. The assay sample and buffer are incubated together with BCAT2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BCAT2 concentration since BCAT2 from samples and BCAT2-HRP conjugate compete for the anti-BCAT2 antibody binding site. Since the number of sites is limited, as more sites are occupied by BCAT2 from the sample, fewer sites are left to bind BCAT2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BCAT2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a C2H2-type zinc finger protein and is closely related to BCL11A, a gene whose translocation may be associated with B-cell malignancies. Although the specific function of this gene has not been determined, the encoded protein is known to be a transcriptional repressor, and is regulated by the NURD nucleosome remodeling and histone deacetylase complex. Four alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Aug 2013]

Uniprot Description

Bcl-11B: Tumor-suppressor protein involved in T-cell lymphomas. May function on the P53-signaling pathway. May be a key regulator of both differentiation and survival during thymocyte development. Repress transcription through direct, TFCOUP2-independent binding to a GC-rich response element. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: C2H2-type zinc finger protein; Nuclear receptor co-regulator

Chromosomal Location of Human Ortholog: 14q32.2

Cellular Component: nucleus

Molecular Function: protein binding; metal ion binding

Biological Process: positive T cell selection; post-embryonic camera-type eye development; regulation of lipid metabolic process; thymus development; transcription, DNA-dependent; regulation of neuron differentiation; T cell differentiation in the thymus; alpha-beta T cell differentiation; odontogenesis of dentine-containing teeth; negative regulation of cell proliferation; striatal medium spiny neuron differentiation; commitment of a neuronal cell to a specific type of neuron in the forebrain; T cell receptor V(D)J recombination; positive regulation of transcription from RNA polymerase II promoter; negative regulation of apoptosis

Product Notes

The Human BCL11B bcl11b (Catalog #AAA7203011) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7203011 ELISA Kit recognizes Human BCL11B. It is sometimes possible for the material contained within the vial of "B cell lymphoma/leukemia 11B (BCL11B), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
