Intended Uses: This DDX3Y ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human DDX3Y. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: DDX3Y ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-DDX3Y antibody and an DDX3Y-HRP conjugate. The assay sample and buffer are incubated together with DDX3Y-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the DDX3Y concentration since DDX3Y from samples and DDX3Y-HRP conjugate compete for the anti-DDX3Y antibody binding site. Since the number of sites is limited, as more sites are occupied by DDX3Y from the sample, fewer sites are left to bind DDX3Y-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The DDX3Y concentration in each sample is interpolated from this standard curve.

NCBI Description

DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD), are putative RNA helicases. They are implicated in a number of cellular processes involving alteration of RNA secondary structure such as translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Based on their distribution patterns, some members of this family are believed to be involved in embryogenesis, spermatogenesis, and cellular growth and division. This gene encodes a DEAD box protein, and it has a homolog on the X chromosome. The gene mutation causes male infertility, Sertoli cell-only syndrome or severe hypospermatogenesis, suggesting that this gene plays a key role in the spermatogenic process.
