Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human Arg antibody. Arg present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human Arg Antibody is added and binds to Arg in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated Arg antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human Arg. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Arginase catalyzes the hydrolysis of arginine to ornithine and urea. At least two isoforms of mammalian arginase exist (types I and II) which differ in their tissue distribution, subcellular localization, immunologic crossreactivity and physiologic function. The type I isoform encoded by this gene, is a cytosolic enzyme and expressed predominantly in the liver as a component of the urea cycle. Inherited deficiency of this enzyme results in argininemia, an autosomal recessive disorder characterized by hyperammonemia.
