Intended Uses: This BAMBI ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Sheep BAMBI. This ELISA kit for research use only!

Principle of the Assay: BAMBI ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-BAMBI antibody and an BAMBI-HRP conjugate. The assay sample and buffer are incubated together with BAMBI-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BAMBI concentration since BAMBI from samples and BAMBI-HRP conjugate compete for the anti-BAMBI antibody binding site. Since the number of sites is limited, as more sites are occupied by BAMBI from the sample, fewer sites are left to bind BAMBI-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BAMBI concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the BCL2 protein family. BCL2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. This protein forms a heterodimer with BCL2, and functions as an apoptotic activator. This protein is reported to interact with, and increase the opening of, the mitochondrial voltage-dependent anion channel (VDAC), which leads to the loss in membrane potential and the release of cytochrome c. The expression of this gene is regulated by the tumor suppressor P53 and has been shown to be involved in P53-mediated apoptosis. Multiple alternatively spliced transcript variants, which encode different isoforms, have been reported for this gene. [provided by RefSeq, Jul 2008]

Accelerates programmed cell death by binding to, and antagonizing the apoptosis repressor BCL2 or its adenovirus homolog E1B 19k protein. Under stress conditions, undergoes a conformation change that causes translocation to the mitochondrion membrane, leading to the release of cytochrome c that then triggers apoptosis. Promotes activation of CASP3, and thereby apoptosis.
