Principle of the Assay: AZIN1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-AZIN1 antibody and an AZIN1-HRP conjugate. The assay sample and buffer are incubated together with AZIN1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AZIN1 concentration since AZIN1 from samples and AZIN1-HRP conjugate compete for the anti-AZIN1 antibody binding site. Since the number of sites is limited, as more sites are occupied by AZIN1 from the sample, fewer sites are left to bind AZIN1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AZIN1 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene belongs to the antizyme inhibitor family, which plays a role in cell growth and proliferation by maintaining polyamine homeostasis within the cell. Antizyme inhibitors are homologs of ornithine decarboxylase (ODC, the key enzyme in polyamine biosynthesis) that have lost the ability to decarboxylase ornithine; however, retain the ability to bind to antizymes. Antizymes negatively regulate intracellular polyamine levels by binding to ODC and targeting it for degradation, as well as by inhibiting polyamine uptake. Antizyme inhibitors function as positive regulators of polyamine levels by sequestering antizymes and neutralizing their effect. This gene encodes antizyme inhibitor 1, the first member of this gene family that is ubiquitously expressed, and is localized in the nucleus and cytoplasm. Overexpression of antizyme inhibitor 1 gene has been associated with increased proliferation, cellular transformation and tumorigenesis. Gene knockout studies showed that homozygous mutant mice lacking functional antizyme inhibitor 1 gene died at birth with abnormal liver morphology.
