Test Principle
This assay employs a two-site sandwich ELISA to quantitate ANXA1 in samples. An antibody specific for ANXA1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyANXA1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ANXA1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ANXA1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Preparation and Storage
Store at 2-8 degree C. Please refer to Instruction Manual.

NCBI Description

Annexin I belongs to a family of Ca(2+)-dependent phospholipid binding proteins which have a molecular weight of approximately 35,000 to 40,000 and are preferentially located on the cytosolic face of the plasma membrane. Annexin I protein has an apparent relative molecular mass of 40 kDa, with phospholipase A2 inhibitory activity. Since phospholipase A2 is required for the biosynthesis of the potent mediators of inflammation, prostaglandins and leukotrienes, annexin I may have potential anti-inflammatory activity. [provided by RefSeq, Jul 2008]

Uniprot Description

ANXA1: a calcium/phospholipid-binding protein with which promotes membrane fusion and is involved in endocytosis. Has anti-inflammatory properties and inhibits phospholipase A2 activity. Accumulates on internalized vesicles after EGF-stimulated endocytosis, suggesting that it may be required for a late stage in inward vesiculation. Phosphorylated by PKC, EGFR and Chak1. Phosphorylation results in loss of the inhibitory activity. Annexins are a family of structurally related proteins whose common property is calcium-dependent binding to phospholipids. There are at least ten different annexins in mammalian species. Annexins do not contain signal peptides, yet some annexins (A1, A2 and A5) appear to be secreted in a physiologically regulated fashion.

Product Notes

The Human ANXA1 anxa1 (Catalog #AAA2802279) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2802279 ELISA Kit recognizes Human ANXA1. It is sometimes possible for the material contained within the vial of "Annexin A1 (ANXA1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
