Background: Angiogenin (ANG) also known as ribonuclease 5, is a potent stimulator of new blood vessel formation. ANG is a member of the pancreatic ribonuclease A superfamily, and RNase activity of ANG is important for its angiogenic activity. It is expressed in the neuroaxis. Like VEGF, ANG is induced by hypoxia to elicit angiogenesis and is expressed in motor neurons. Angiogenin may function as a tRNA-specific ribonuclease that binds to actin on the surface of endothelial cells, once bound, angiogenin is endocytosed and translocated to the nucleus, thereby promoting the endothelial invasiveness necessary for blood vessel formation. Angiogenin induces vascularization of normal and malignant tissues, and abolishes protein synthesis by specifically hydrolyzing cellular tRNAs.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is an exceedingly potent mediator of new blood vessel formation. It hydrolyzes cellular tRNAs resulting in decreased protein synthesis and is similar to pancreatic ribonuclease. Alternative splicing results in two transcript variants encoding the same protein. This gene and the gene that encodes ribonuclease, RNase A family, 4 share promoters and 5' exons. Each gene splices to a unique downstream exon that contains its complete coding region. Uniprot Description
Function: Binds to actin on the surface of endothelial cells; once bound, angiogenin is endocytosed and translocated to the nucleus. Stimulates ribosomal RNA synthesis including that containing the initiation site sequences of 45S rRNA. Cleaves tRNA within anticodon loops to produce tRNA-derived stress-induced fragments (tiRNAs) which inhibit protein synthesis and triggers the assembly of stress granules (SGs). Angiogenin induces vascularization of normal and malignant tissues. Angiogenic activity is regulated by interaction with RNH1 in vivo. Ref.10 Ref.14 Ref.15 Ref.16
Subunit structure: Interacts with and forms a tight 1:1 complex with RNH1. Dimerization of two such complexes may occur. Ref.13 Ref.15 Ref.18
Subcellular location: Secreted › extracellular space › extracellular matrix › basement membrane. Nucleus › nucleolus. Note: Rapidly endocytosed by target cells and translocated to the nucleus where it accumulates in the nucleolus and binds to DNA. Ref.11 Ref.14
Tissue specificity: Expressed predominantly in the liver. Also detected in endothelial cells and spinal cord neurons. Ref.9 Ref.28
Developmental stage: Low level expression in the developing fetus, increased in the neonate, and maximal in the adult.
Involvement in disease: Amyotrophic lateral sclerosis 9 (ALS9) [MIM:611895]: A neurodegenerative disorder affecting upper motor neurons in the brain and lower motor neurons in the brain stem and spinal cord, resulting in fatal paralysis. Sensory abnormalities are absent. The pathologic hallmarks of the disease include pallor of the corticospinal tract due to loss of motor neurons, presence of ubiquitin-positive inclusions within surviving motor neurons, and deposition of pathologic aggregates. The etiology of amyotrophic lateral sclerosis is likely to be multifactorial, involving both genetic and environmental factors. The disease is inherited in 5-10% of the cases.Note: Disease susceptibility is associated with variations affecting the gene represented in this entry. Ref.26 Ref.27 Ref.28 Ref.29 Ref.30 Ref.31
Sequence similarities: Belongs to the pancreatic ribonuclease family.
Caution: It is uncertain whether Met-1 or Met-3 is the initiator.
Sequence caution: The sequence AAH20704.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended.

Product Notes
The Human ANG ang (Catalog #AAA355287) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355287 ELISA Kit recognizes Human ANG. AAA Biotech's Angiogenin / ANG can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of ANG in human serum, plasma, body fluids, tissue lysates or cell culture supernatants. Researchers should empirically determine the suitability of the ANG ang for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Angiogenin / ANG, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
