Human
Specificity
This assay has high sensitivity and excellent specificity for detection of ADRA2B. No significant cross-reactivity or interference between ADRA2B and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ADRA2B and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This ADRA2B ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ADRA2B. This ELISA kit for research use only!

Principle of the Assay: ADRA2B ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ADRA2B antibody and an ADRA2B-HRP conjugate. The assay sample and buffer are incubated together with ADRA2B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADRA2B concentration since ADRA2B from samples and ADRA2B-HRP conjugate compete for the anti-ADRA2B antibody binding site. Since the number of sites is limited, as more sites are occupied by ADRA2B from the sample, fewer sites are left to bind ADRA2B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADRA2B concentration in each sample is interpolated from this standard curve.

NCBI Description

This intronless gene encodes a seven-pass transmembrane protein. This protein is a member of a subfamily of G protein-coupled receptors that regulate neurotransmitter release from sympathetic nerves and from adrenergic neurons in the central nervous system. [provided by RefSeq, Apr 2014]

Uniprot Description

ADRA2B: Alpha-2 adrenergic receptors mediate the catecholamine- induced inhibition of adenylate cyclase through the action of G proteins. The rank order of potency for agonists of this receptor is clonidine > norepinephrine > epinephrine = oxymetazoline > dopamine > p-tyramine = phenylephrine > serotonin > p-synephrine / p-octopamine. For antagonists, the rank order is yohimbine > chlorpromazine > phentolamine > mianserine > spiperone > prazosin > alprenolol > propanolol > pindolol. Belongs to the G-protein coupled receptor 1 family. Adrenergic receptor subfamily. ADRA2B sub-subfamily.

Protein type: GPCR, family 1; Receptor, GPCR; Membrane protein, integral; Membrane protein, multi-pass

Chromosomal Location of Human Ortholog: 2q11.1

Cellular Component: integral to plasma membrane; plasma membrane

Molecular Function: protein binding; alpha2-adrenergic receptor activity; epinephrine binding

Biological Process: G-protein coupled receptor protein signaling pathway; platelet activation; cell-cell signaling; activation of protein kinase B; regulation of vascular smooth muscle contraction; negative regulation of epinephrine secretion; MAPKKK cascade; female pregnancy; angiogenesis; positive regulation of neuron differentiation; blood coagulation; positive regulation of blood pressure

Product Notes

The Human ADRA2B adra2b (Catalog #AAA7246520) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7246520 ELISA Kit recognizes Human ADRA2B. It is sometimes possible for the material contained within the vial of "Alpha 2B adrenergic receptor (ADRA2B), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
