Intended Uses: This MGAT5 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human MGAT5. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: MGAT5 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-MGAT5 antibody and an MGAT5-HRP conjugate. The assay sample and buffer are incubated together with MGAT5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MGAT5 concentration since MGAT5 from samples and MGAT5-HRP conjugate compete for the anti-MGAT5 antibody binding site. Since the number of sites is limited, as more sites are occupied by MGAT5 from the sample, fewer sites are left to bind MGAT5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MGAT5 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the glycosyltransferase family. It catalyzes the addition of beta-1,6-N-acetylglucosamine to the alpha-linked mannose of biantennary N-linked oligosaccharides present on the newly synthesized glycoproteins. It is one of the most important enzymes involved in the regulation of the biosynthesis of glycoprotein oligosaccharides. Alterations of the oligosaccharides on cell surface glycoproteins cause significant changes in the adhesive or migratory behavior of a cell. Increase in the activity of this enzyme has been correlated with the progression of invasive malignancies. [provided by RefSeq, Oct 2011]
