Intended Uses: This sandwich kit is for the accurate quantitative detection of human Aflatoxin B1 Aldehyde Reductase Member 4 (also known as AKR7L) in serum, plasma, cell culture supernates, cell lysates, tissue homogenates.

Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with human AKR7L antibody. AKR7L present in the sample is added and binds to antibodies coated on the wells. And then biotinylated human AKR7L Antibody is added and binds to AKR7L in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated AKR7L antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of human AKR7L. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

NCBI Description

This gene is one of three aldo-keto reductase genes that are present in a cluster on the p arm of chromosome 1. The encoded proteins are involved in the reduction of the dialdehyde protein-binding form of aflatoxin B1 (AFB1) to the non-binding AFB1 dialcohol. It has been speculated that this family member encodes a selenoprotein, which includes a selenocysteine (Sec) residue in lieu of a UGA translational termination codon. However, there is no evidence that such a protein is produced in vivo. The alternative interpretation is that this family member is a segregating pseudogene, where some individuals have an allele that encodes a functional enzyme, while other individuals have an allele encoding a protein that is predicted to be non-functional.
