Human Aflatoxin B1 aldehyde reductase member 2 (AKR7A2) ELISA Kit
Principle of the Assay: AKR7A2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-AKR7A2 antibody and an AKR7A2-HRP conjugate. The assay sample and buffer are incubated together with AKR7A2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AKR7A2 concentration since AKR7A2 from samples and AKR7A2-HRP conjugate compete for the anti-AKR7A2 antibody binding site. Since the number of sites is limited, as more sites are occupied by AKR7A2 from the sample, fewer sites are left to bind AKR7A2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AKR7A2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene belongs to the aldo/keto reductase (AKR) superfamily and AKR7 family, which are involved in the detoxification of aldehydes and ketones. The AKR7 family consists of 3 genes that are present in a cluster on the p arm of chromosome 1. This protein, thought to be localized in the golgi, catalyzes the NADPH-dependent reduction of succinic semialdehyde to the endogenous neuromodulator, gamma-hydroxybutyrate. It may also function as a detoxication enzyme in the reduction of aflatoxin B1 and 2-carboxybenzaldehyde. [provided by RefSeq, Oct 2011]

Can reduce 1,2-naphthoquinone and 9,10-phenanthrenequinone (in vitro). Can reduce the dialdehyde protein-binding form of aflatoxin B1 (AFB1) to the non-binding AFB1 dialcohol.
