Principle of the Assay: MLLT4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-MLLT4 antibody and an MLLT4-HRP conjugate. The assay sample and buffer are incubated together with MLLT4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MLLT4 concentration since MLLT4 from samples and MLLT4-HRP conjugate compete for the anti-MLLT4 antibody binding site. Since the number of sites is limited, as more sites are occupied by MLLT4 from the sample, fewer sites are left to bind MLLT4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MLLT4 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a multi-domain protein involved in signaling and organization of cell junctions during embryogenesis. It has also been identified as the fusion partner of acute lymphoblastic leukemia (ALL-1) gene, involved in acute myeloid leukemias with t(6;11)(q27;q23) translocation. Alternatively spliced transcript variants encoding different isoforms have been described for this gene, however, not all have been fully characterized.[provided by RefSeq, May 2011]
Uniprot Description
Afadin: plays a role in adhesion by participating, along with E-cadherin and catenin, in the organization of homotypic, interneuronal and heterotypic cell-cell adherens junctions (AJs). Essential for the association of nectin and E-cadherin. Contains 1 forkhead-associated (FHA) domain, a phosphopeptide recognition domain; 1 dilute (DIL) domain, a myosin-like domain which may play a functional role in cellular processes or vectorial vesicle transport; 1 PDZ domain, which frequently binds the carboxyl-terminal sequences of proteins or phosphatidylinositol 4,5-bisphosphate in the plasma membrane; and 2 RA domains, which are associated with RasGTP effector activity. Five alternatively spliced isoforms have been described. Isoform 2 does not interact with F-actin.

Product Notes
The Human MLLT4 mllt4 (Catalog #AAA7214285) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7214285 ELISA Kit recognizes Human MLLT4. It is sometimes possible for the material contained within the vial of "Afadin (MLLT4), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
