Sequence
The extracellular domain of human TRAIL (aa 95-281) is fused at the N-terminus to a His-tag and a linker peptide and contains aa substitutions, which generate a binding specificity towards TRAIL receptor 1 (TRAIL-R1; DR4).
Biological Activity
Induces apoptosis in a concentration range of 20-500ng/ml of TRAIL-R1 (DR4) expressing cell lines (e.g. RAMOS). It is inefficient to kill via TRAIL-R2 (DR5) in Jurkat cells. The activity depends on the relative cellular surface expression of TRAIL-R1 compared to other death-signaling TRAIL-Rs. Concentrations of KillerTRAIL-R1 to induce apoptosis may vary depending on the cell type.

Note
This protein does not require a cross-linking enhancer for its potent biological activity.

NCBI Description

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor is activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL), and thus transduces cell death signal and induces cell apoptosis. Studies with FADD-deficient mice suggested that FADD, a death domain containing adaptor protein, is required for the apoptosis mediated by this protein. [provided by RefSeq, Jul 2008]
