Assay Procedure Summary
1. Prepare all reagents, samples and standards
2. Add 100ul standard or sample to each well. Incubate 2 hours at 37 degree C
3. Aspirate and add 100ul prepared Detection Reagent A. Incubate 1 hour at 37 degree C
4. Aspirate and wash 3 times
5. Add 100ul prepared Detection Reagent B. Incubate 30 minutes at 37 degree C
6. Aspirate and wash 5 times
7. Add 100ul Substrate Solution. Incubate 10 minutes at 37 degree C
8. Read RLU value immediately.
Test Principle
The microplate provided in this kit has been pre-coated with an antibody specific to X-Ray Repair Cross Complementing 6 (XRCC6). Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to X-Ray Repair Cross Complementing 6 (XRCC6). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is proportional to the X-Ray Repair Cross Complementing 6 (XRCC6) level in the sample or standard.;
Preparation and Storage
Store at 4 degree C for 1 month/ -20 degree C for long-term (within shelf life).
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

NCBI Description

The protein encoded by this gene is a GTPase which belongs to the RAS superfamily of small GTP-binding proteins. Members of this superfamily appear to regulate a diverse array of cellular events, including the control of cell growth, cytoskeletal reorganization, and the activation of protein kinases. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2009]

Uniprot Description

RAC1: a plasma membrane-associated member of the Rho-GTPase family. Plays a key role in cytoskeletal reorganization, membrane trafficking, transcriptional regulation and cell growth and development. GTP binding stimulates its activity. Phosphorylation by Akt may inhibit GTP binding of Rac1, therefore attenuating the downstream signal transduction pathway. Found in a trimeric complex composed of DOCK1 and ELMO1, which plays a central role in phagocytosis of apoptotic cells. Two alternatively spliced isoforms have been described.
