Intended Uses: This CLIA kit applies to the in vitro quantitative determination of PLD2 concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This kit uses Sandwich-CLIA as the method. The micro CLIA plate provided in this kit has been pre-coated with an antibody specific to PLD2. Standards or samples are added to the appropriate micro CLIA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for PLD2 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each micro plate well successively and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain PLD2, biotinylated detection antibody and Avidin-HRP conjugate will appear fluorescence. The Relative light unit (RLU) value is measured spectrophotometrically by the Chemiluminescence immunoassay analyzer. The RLU value is positively associated with the concentration of PLD2. You can calculate the concentration of PLD2 in the samples by comparing the RLU value of the samples to the standard curve.

NCBI Description

The protein encoded by this gene catalyzes the hydrolysis of phosphatidylcholine to phosphatidic acid and choline. The activity of the encoded enzyme is enhanced by phosphatidylinositol 4,5-bisphosphate and ADP-ribosylation factor-1. This protein localizes to the peripheral membrane and may be involved in cytoskeletal organization, cell cycle control, transcriptional regulation, and/or regulated secretion. Two transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Jul 2011]

PLD2: a phosphatidylcholine-specific phospholipase D. Catalyzes the hydrolysis of PC to produce phosphatidic acid and choline. Activated as a consequence of agonist stimulation of both tyrosine kinase and G protein-coupled receptors. May function in regulated secretion, cytoskeletal reorganization, transcriptional regulation, and cell cycle control. Enriched in caveolae; may be involved in MEK/ERK signaling cascades induced by the VEGF/VEGFR-2/PKC-delta pathway in endothelial cells. Three alternatively spliced isoforms have been described.
