Principle of the Assay: The LumiAb Human MIP-1alpha Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human MIP-1alpha concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human MIP-1alpha while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human MIP-1alpha or Macrophage Inflammatory Protein-1alpha, also known as Chemokine (C-C Motif) Ligand 3, is a 92 amino acid inducible chemokine expressed from the gene CCL3 located at locus 17q12 on chromosome 17. After initial translation of the protein, the peptide is proteolytically cleaved to separate the 23 residue signal sequence from the 69 residue MIP-1alpha mature chemokine. Structurally, the small protein is composed of 2 beta strands of 3 residues each and a 6 residue helix towards the C-terminus of the peptide. MIP-1alpha is a monokine that primarily plays a role in inflammatory responses through binding to the CCR1, CCR4 and CCR5 receptors. This small protein is one of the major HIV-suppressive factors produced by CD8+ T-cells such that it can induce a dose-dependent inhibition of several different strains of HIV-1, HIV-2 and simian immunodeficiency virus (SIV). MIP-1alpha is inducible by the tumor promoter 12-O-tetradecanoyl phorbol-13 acetate (TPA) or the T-cell mitogen phytohemagglutinin (PHA). Along with its counterpart, MIP-1beta, these factors can activate human granulocytes (neutrophils, eosinophils and basophils) as well as induce the synthesis and release of other pro-inflammatory cytokines such as IL-1, IL-6 and TNF-alpha from fibroblasts and macrophages.

The protein encoded by this gene is a platelet-derived growth factor that belongs to the CXC chemokine family. This growth factor is a potent chemoattractant and activator of neutrophils. It has been shown to stimulate various cellular processes including DNA synthesis, mitosis, glycolysis, intracellular cAMP accumulation, prostaglandin E2 secretion, and synthesis of hyaluronic acid and sulfated glycosaminoglycan. It also stimulates the formation and secretion of plasminogen activator by synovial cells. The protein also is an antimicrobial protein with bactericidal and antifungal activity. PPBP: LA-PF4 stimulates DNA synthesis, mitosis, glycolysis, intracellular cAMP accumulation, prostaglandin E2 secretion, and synthesis of hyaluronic acid and sulfated glycosaminoglycan. It also stimulates the formation and secretion of plasminogen activator by human synovial cells. NAP-2 is a ligand for CXCR1 and CXCR2, and NAP-2, NAP-2(73), NAP-2(74), NAP-2(1-66), and most potent NAP-2(1-63) are chemoattractants and activators for neutrophils. TC-1 and TC-2 are antibacterial proteins, in vitro released from activated platelet alpha-granules. CTAP-III(1-81) is more potent than CTAP-III desensitize chemokine-induced neutrophil activation. Belongs to the intercrine alpha (chemokine CxC) family.
