Specific Activity
20 U/ug. Sphingosine kinase activity was determined using D- erythro-sphingosine and [ -32P]ATP as substrates at pH 7.4 and 30 degree C. A unit of sphingosine kinase activity is defined as the amount of enzyme required to produce 1pmol of S1P/min.

50mM HEPES, pH 7.4, 150mM NaCl, 5mM MgCl2, 1mM DTT, 3uM Na-orthovanadate, 0.5mM ATP, 4uM D-erythro-sphingosine, and 1ug/ml sphingosine kinase 2.

NCBI Description

This gene encodes one of two sphingosine kinase isozymes that catalyze the phosphorylation of sphingosine into sphingosine 1-phosphate. Sphingosine 1-phosphate mediates many cellular processes including migration, proliferation and apoptosis, and also plays a role in several types of cancer by promoting angiogenesis and tumorigenesis. The encoded protein may play a role in breast cancer proliferation and chemoresistance. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. SPHK2: Catalyzes the phosphorylation of sphingosine to form sphingosine 1-phosphate (SPP), a lipid mediator with both intra- and extracellular functions. Also acts on D-erythro- dihydrosphingosine, D-erythro-sphingosine and L-threo- dihydrosphingosine. Binds phosphoinositides. 3 isoforms of the human protein are produced by alternative splicing.
