Biological Significance
Follistatin is a secreted protein that binds to ligands of the TGF beta family and regulates their activity by inhibiting their access to signaling receptors. It was originally discovered as activin antagonists whose activity suppresses expression and secretion of the pituitary hormone FSH (follicle stimulating hormone). In addition to being a natural antagonist, follistatin can inhibit the activity of other TGF beta ligands including BMP-2, -4, -6, -7, Myostatin, GDF-11, and TGF beta1. Follistatin is expressed in the pituitary, ovaries, decidual cells of the endometrium, and in some other tissues.
Bioactivity
Determined by its ability to neutralize Activin A inhibitory effect of Mouse MPC-11 cells. The expected ED50 is 0.1-0.4 ug/ml in the presence of 7.5 ng/ml Activin A.
Endotoxin Levels
Endotoxin level is less than 0.1 ng per ug (1 EU/ug).
Expression System
E Coli
Preparation and Storage
Store at 4 degree C until reconstitution. Following reconstitution aliquot and freeze at -20 degree C for long term storage. Avoid repeated freeze/thaw cycles.

NCBI Description

The protein encoded by this gene binds to and negatively regulates activin, as well as other members of the transforming growth factor beta family, and acts to prevent uncontrolled cellular proliferation. This protein also contains a heparin-binding sequence. It is expressed in many of the tissues in which activin is synthesized and is thought to clear activin from the circulation by attachment to the cell surface. Alternative splicing results in multiple transcript variants that encode multiple protein isoforms, including FST315 and FST288, that differ at their C-terminus. Another isoform, FST303 is thought to be produced by proteolytic cleavage of FST315. These isoforms differ in their localization and in their ability to bind heparin. While FST315 is a circulating protein, FST288 is tissue-bound, and FST303 is gonad-specific. While deletion of all isoforms results in embryonic lethality, expression of just FST288 is sufficient for embryonic development, but the resultant mice have fertility defects. Product Notes

The Fst fst (Catalog #AAA538118) is an Active Protein produced from Human and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Follistatin can be used in a range of immunoassay formats including, but not limited to, User optimized. Researchers should empirically determine the suitability of the Fst fst for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: Region of Follistati n protein correspond ing to amino acids GNCWLRQAKN GRCQVLYKTE LSKEECCSTG RLSTSWTEED VNDNTLFKWM IFNGGAPNCI PCKETCENVD CGPGKKCRMN KKNKPRCVCA PDCSNITWKG PVCGLDGKTY RNECALLKAR CKEQPELEVQ YQGRCKKTCR DVFCPGSSTC VVDQTNNAYC VTCNRICPEP ASSEQYLCGN DGVTYSSACH LRKATCLLGR SIGLAYEGKC IKAKSCEDIQ CTGGKKCLWD FKVGRGRCSL CDELCPDSKS DEPVCASDNA TYASECAMKE AACSSGVLLE VKHSGSCN. It is sometimes possible for the material contained within the vial of "Follistatin, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
