Biological Significance
Defensins (alpha and beta) are cationic peptides with a broad spectrum of antimicrobial activity that comprise an important arm of the innate immune system. The a-defensins are distinguished from the beta-defensins by the pairing of their three disulfide bonds. To date, four human beta-defensins have been identified; BD-1, BD-2, BD-3 and BD-4. The beta-defensin proteins are expressed as the C-terminal portion of precursors and are released by proteolytic cleavage of a signal sequence. beta-defensins contain a six-cysteine motif that forms three intra-molecular disulfide bonds. beta-defensins are 3-5 kDa peptides ranging in size from 33-47 amino acid residues. BD-4 is expressed in testis, stomach, uterus, neutrophils, thyroid, lung and kidney.
Bioactivity
Determined by its ability to chemoattract human monocytes using a concentration range of 0.1-100.0 ng/ml.
Endotoxin
Endotoxin level is <0.1 ng/ug of protein (1 EU/ug).
Expression System
E Coli
Form
Sterile filtered through a 0.2 micron filter. Lyophilized from 10mM Acetic Acid. Initially reconstitute in 10mM Acetic Acid to 0.1-1.0mg/mL. After initial reconstitution , further dilute in a buffer containing a carrier protein or stabilizer (e.g. 0.1% BSA)
Preparation and Storage
Store at 4 degree C until reconstitution. Following reconstitution aliquot and freeze at -20 degree C for long term storage. Avoid repeated freeze/thaw cycles.

NCBI Description

Defensins form a family of antimicrobial and cytotoxic peptides made by neutrophils. Defensins are short, processed peptide molecules that are classified by structure into three groups: alpha-defensins, beta-defensins and theta-defensins. All beta-defensin genes are densely clustered in four to five syntenic chromosomal regions. Chromosome 8p23 contains at least two copies of the duplicated beta-defensin cluster. This duplication results in two identical copies of defensin, beta 104, DEFB104A and DEFB104B, in head-to-head orientation. This gene, DEFB104A, represents the more centromeric copy. Product Notes

The BD4 defb104a (Catalog #AAA539480) is an Active Protein produced from Human and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's BD4 can be used in a range of immunoassay formats including, but not limited to, User optimized. Researchers should empirically determine the suitability of the BD4 defb104a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: Region of BD4 protein correspond ing to amino acids EFELDRICGY GTARCRKKCR SQEYRIGRCP NTYACCLRKW DESLLNRTKP. It is sometimes possible for the material contained within the vial of "BD4, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
