Principle of the assay: HTN5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HTN5 antibody and an HTN5-HRP conjugate. The assay sample and buffer are incubated together with HTN5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HTN5 concentration since HTN5 from samples and HTN5-HRP conjugate compete for the anti-HTN5 antibody binding site. Since the number of sites is limited, as more sites are occupied by HTN5 from the sample, fewer sites are left to bind HTN5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HTN5 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the histatin family of small, histidine-rich, cationic proteins. They function as antimicrobial peptides and are important components of the innate immune system. Histatins are found in saliva and exhibit antibacterial, antifungal activities and function in wound healing. [provided by RefSeq, Sep 2014]

HTN3: Histatins are salivary proteins that are considered to be major precursors of the protective proteinaceous structure on tooth surfaces (enamel pellicle). In addition, histatins exhibit antibacterial and antifungal activities. His3-(20-43)-peptide (histatin-5) is especially effective against C.albicans and C.neoformans, and inhibits Lys-gingipain and Arg-gingipain (rgpB) from P.gingivalis. In addition, His3-(20-43)-peptide is a potent inhibitor of metalloproteinases MMP2 and MMP9. Belongs to the histatin/statherin family.
