Principle of the assay: HSL ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-HSL antibody and an HSL-HRP conjugate. The assay sample and buffer are incubated together with HSL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HSL concentration since HSL from samples and HSL-HRP conjugate compete for the anti-HSL antibody binding site. Since the number of sites is limited, as more sites are occupied by HSL from the sample, fewer sites are left to bind HSL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HSL concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene has a long and a short form, generated by use of alternative translational start codons. The long form is expressed in steroidogenic tissues such as testis, where it converts cholesteryl esters to free cholesterol for steroid hormone production. The short form is expressed in adipose tissue, among others, where it hydrolyzes stored triglycerides to free fatty acids. [provided by RefSeq, Jul 2008]

HSL: hormone sensitive lipase is a lipolytic enzyme of the 'GDXG' family. Plays a rate limiting step in triglyceride lipolysis. In adipose tissue and heart, it primarily hydrolyzes stored triglycerides to free fatty acids, while in steroidogenic tissues, it principally converts cholesteryl esters to free cholesterol for steroid hormone production. Rapidly activated by cAMP-dependent phosphorylation under the influence of catecholamines. Dephosphorylation and inactivation are controlled by insulin.
