Reactivity
Human
Synonyms
Heparanase; Human Heparanase ELISA Kit; HPA elisa kit
Ordering
For Research Use Only!
Reactivity
Human
Specificity
No significant cross-reactivity or interference between Human HPA and analogues was observed.
Samples
Serum, Plasma, Tissue Homogenate, Feces, Urine and Body Fluids
Assay Type
Sandwich
Detection Range
6.25 U/ml - 200 U/ml.
Sensitivity
1.0 U/ml.
Intended Uses
This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic applications! It is intended to be determinated HPA concentrations in Human serum, plasma and other body fluids. Using Purified Human HPA antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add HPA and HPA antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of HPA in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Intra-assay Precision
Intra-assay CV (%) is less than 15%.
Inter-assay Precision
Inter-assay CV (%) is less than 15%. [CV(%) = SD/mean x100]
Preparation and Storage
Store all reagents at 2-8 degree C

NCBI Description

Heparan sulfate proteoglycans are major components of the basement membrane and extracellular matrix. The protein encoded by this gene is an enzyme that cleaves heparan sulfate proteoglycans to permit cell movement through remodeling of the extracellular matrix. In addition, this cleavage can release bioactive molecules from the extracellular matrix. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2011]

Uniprot Description

HPSE: Endoglycosidase that cleaves heparan sulfate proteoglycans (HSPGs) into heparan sulfate side chains and core proteoglycans. Participates in extracellular matrix (ECM) degradation and remodeling. Selectively cleaves the linkage between a glucuronic acid unit and an N-sulfo glucosamine unit carrying either a 3-O-sulfo or a 6-O-sulfo group. Can also cleave the linkage between a glucuronic acid unit and an N-sulfo glucosamine unit carrying a 2-O-sulfo group, but not linkages between a glucuronic acid unit and a 2-O-sulfated iduronic acid moiety. It is essentially inactive at neutral pH but becomes active under acidic conditions such as during tumor invasion and in inflammatory processes. Facilitates cell migration associated with metastasis, wound healing and inflammation. Enhances shedding of syndecans, and increases endothelial invasion and angiogenesis in myelomas. Acts as procoagulant by increasing the generation of activation factor X in the presence of tissue factor and activation factor VII. Increases cell adhesion to the extacellular matrix (ECM), independent of its enzymatic activity. Induces AKT1/PKB phosphorylation via lipid rafts increasing cell mobility and invasion. Heparin increases this AKT1/PKB activation. Regulates osteogenesis. Enhances angiogenesis through up- regulation of SRC-mediated activation of VEGF. Implicated in hair follicle inner root sheath differentiation and hair homeostasis. Belongs to the glycosyl hydrolase 79 family.

Product Notes

The Human HPA hpse (Catalog #AAA041301) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA041301 ELISA Kit recognizes Human HPA. It is sometimes possible for the material contained within the vial of "Heparanase, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
