Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- TAFI antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-TAFI antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the TAFI amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of TAFI can be calculated.

NCBI Description

This gene encodes carboxypeptidase B, a zinc-dependent metalloprotease that cleaves peptide bonds at the C-terminus of protein substrates. The encoded preproprotein undergoes proteolytic activation to generate a mature, functional enzyme, and secreted into plasma. [provided by RefSeq, Jan 2016]

CPB2: Cleaves C-terminal arginine or lysine residues from biologically active peptides such as kinins or anaphylatoxins in the circulation thereby regulating their activities. Down- regulates fibrinolysis by removing C-terminal lysine residues from fibrin that has already been partially degraded by plasmin. Belongs to the peptidase M14 family. 2 isoforms of the human protein are produced by alternative splicing.
