Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for HMGB-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HMGB-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HMGB-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HMGB-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a protein that belongs to the High Mobility Group-box superfamily. The encoded non-histone, nuclear DNA-binding protein regulates transcription, and is involved in organization of DNA. This protein plays a role in several cellular processes, including inflammation, cell differentiation and tumor cell migration. Multiple pseudogenes of this gene have been identified. Alternative splicing results in multiple transcript variants that encode the same protein. [provided by RefSeq, Sep 2015]

Function: DNA binding proteins that associates with chromatin and has the ability to bend DNA. Binds preferentially single-stranded DNA. Involved in V(D)J recombination by acting as a cofactor of the RAG complex. Acts by stimulating cleavage and RAG protein binding at the 23 bp spacer of conserved recombination signal sequences (RSS). Heparin-binding protein that has a role in the extension of neurite-type cytoplasmic processes in developing cells.1 PublicationManual assertion based on experiment in:Ref.7
