Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for HEXIM1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HEXIM1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HEXIM1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HEXIM1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

HEXIM1: Transcriptional regulator which functions as a general RNA polymerase II transcription inhibitor. In cooperation with 7SK snRNA sequesters P-TEFb in a large inactive 7SK snRNP complex preventing RNA polymerase II phosphorylation and subsequent transcriptional elongation. May also regulate NF-kappa-B, ESR1, NR3C1 and CIITA-dependent transcriptional activity. Homooligomer and heterooligomer with HEXIM2; probably dimeric. Component of the 7SK snRNP complex at least composed of P-TEFb (composed of CDK9 and CCNT1/cyclin-T1), HEXIM1, HEXIM2, BCDIN3, SART3 proteins and 7SK and U6 snRNAs.
