The hepatitis B virus is major cause for human liver disease, chronic infection usually causes liver cancer and cirrhosis. The outer viron is covered by a layer derived from endoplasmic reticulum and virus-encoded surface protein. Inside of this layer encloses the viral nucleocapside, i.e. core antigen which contains the viral genome. The capsid consisits of dimmers of a 183-residue protein, which can be divided into a fragment (1-149) responsible for assembling and a basic residue-composed domain responsible for polymerization into particle and RNA packaging, respectively. The core antigen is found on virus particles but disappears early in the course of infection. This antibody is produced during and after an acute HBV infection and is also found in chronic HBV carriers as well as those who have cleared the virus, and usually persists for life. This recombinant core antigen consists of HBV core immunodominant region amino acids 1-183. It was expressed from E Coli, the purified recombinant core antigen can be used for ELISA assay and Western blot for clinical study and research. Some times, virus like particle can be found from over 30% E Coli expressed viral antigen, multiple antigens can aggregate into one large particle, even though with reducing condition, multiplized proteins, such as dimer, trimer is still present on SDS-PAGE. Aggregated virus like particle has strong ability in immunal reaction, they can be used for vaccine study or generate better result in immunoassay for identification.
