Protein Construction
A DNA sequence encoding the rat TNFRSF1A (P22934)(Met1-Ala211) was expressed, fused with the Fc region of human IgG1 at the C-terminus.
Reconstitution
A hardcopy of COA with reconstitution instruction is sent along with the products. Please refer to it for detailed information.
Activity
1. Measured by its ability to inhibit TNFalpha-mediated cytotoxicity in L-929 mouse fibroblast cells in the presence of metabolic inhibitor actinomycin D. The ED50 for this effect is typically 3-30ng/mL in the presence of 0.05ng/mL of ratTNFalpha.
2. Immobilized mouse TNFa (80-235) at 10ug/ml (100uL/well) can bind rat TNFRSF1A-Fc. The EC50 of rat TNFRSF1A-Fc is 0.08-0.20ug/ml.
Preparation and Storage
Samples are stable for up to twelve months from date of receipt at -20 degree C to -80 degree C. Store it under sterile conditions at -20 degree C to -80 degree C. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.
In general, recombinant proteins are provided as lyophilized powder which are shipped at ambient temperature. Bulk packages of recombinant proteins are provided as frozen liquid.
They are shipped out with blue ice.

Related Product Information for TNFRSF1A active protein
Background: The cluster of differentiation (CD) system is commonly used as cell markers in Immunophenotyping. Different kinds of cells in the immune system can be identified through the surface CD molecules which associating with the immune function of the cell. There are more than 320 CD unique clusters and subclusters have been identified. Some of the CD molecules serve as receptors or ligands important to the cell through initiating a signal cascade which then alter the behavior of the cell. Some CD proteins do not take part in cell signal process but have other functions such as cell adhesion. CD120a (cluste of differentiation 120a), also known as TNFR1/TNFRSF1A, is a member of CD family, tumor necrosis factor receptor superfamily. CD120a is one of the most primary receptors for the tumor necrosis factor-alpha. It has been shown to be localized to both plasma membrane lipid rafts and the trans golgi complex with the help of the death domain (DD). CD120a can activate the transcription factor NF-kappaB, mediate apoptosis, and regulate inflammation processes.

NCBI Description

may play a role in TNFalpha mediated inflammatory response to spinal cord injury [RGD, Feb 2006]

Uniprot Description

Receptor for TNFSF2/TNF-alpha and homotrimeric TNFSF1/lymphotoxin-alpha. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis ().

Product Notes

The TNFRSF1A tnfrsf1a (Catalog #AAA8123372) is an Active Protein produced from HEK293 Cells and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: Met1-Ala21 1. It is sometimes possible for the material contained within the vial of "TNFRSF1A, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
