Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-HDAC antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-HDAC antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the HDAC amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of HDAC can be calculated.

NCBI Description

Histones play a critical role in transcriptional regulation, cell cycle progression, and developmental events. Histone acetylation/deacetylation alters chromosome structure and affects transcription factor access to DNA. The protein encoded by this gene belongs to class I of the histone deacetylase family. It catalyzes the deacetylation of lysine residues in the histone N-terminal tails and represses transcription in large multiprotein complexes with transcriptional co-repressors. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2009]

Uniprot Description

HDAC8: Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. May play a role in smooth muscle cell contractility. Belongs to the histone deacetylase family. HD type 1 subfamily. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human HDAC8 hdac8 (Catalog #AAA765934) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA765934 ELISA Kit recognizes Human HDAC8. It is sometimes possible for the material contained within the vial of "Histone deacetylase 8, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
