Principle of the assay: GRN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GRN antibody and an GRN-HRP conjugate. The assay sample and buffer are incubated together with GRN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GRN concentration since GRN from samples and GRN-HRP conjugate compete for the anti-GRN antibody binding site. Since the number of sites is limited, as more sites are occupied by GRN from the sample, fewer sites are left to bind GRN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GRN concentration in each sample is interpolated from this standard curve.

NCBI Description

This antimicrobial gene encodes a member of the CXC subfamily of chemokines. The encoded protein is a secreted growth factor that signals through the G-protein coupled receptor, CXC receptor 2. This protein plays a role in inflammation and as a chemoattractant for neutrophils. Aberrant expression of this protein is associated with the growth and progression of certain tumors. A naturally occurring processed form of this protein has increased chemotactic activity. Alternate splicing results in coding and non-coding variants of this gene. A pseudogene of this gene is found on chromosome 4. [provided by RefSeq, Sep 2014]
