Rat Glutamate Receptor, Ionotropic, Delta 2 ELISA Kit
Principle of the Assay: GRID2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GRID2 antibody and an GRID2-HRP conjugate. The assay sample and buffer are incubated together with GRID2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GRID2 concentration since GRID2 from samples and GRID2-HRP conjugate compete for the anti-GRID2 antibody binding site. Since the number of sites is limited, as more sites are occupied by GRID2 from the sample, fewer sites are left to bind GRID2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GRID2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the family of ionotropic glutamate receptors which are the predominant excitatory neurotransmitter receptors in the mammalian brain. The encoded protein is a multi-pass membrane protein that is expressed selectively in cerebellar Purkinje cells. A point mutation in the mouse ortholog, associated with the phenotype named 'lurcher', in the heterozygous state leads to ataxia resulting from selective, cell-autonomous apoptosis of cerebellar Purkinje cells during postnatal development. Mice homozygous for this mutation die shortly after birth from massive loss of mid- and hindbrain neurons during late embryogenesis. This protein also plays a role in synapse organization between parallel fibers and Purkinje cells. Alternate splicing results in multiple transcript variants encoding distinct isoforms.
