Principle of the assay: GFRBP14 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GFRBP14 antibody and an GFRBP14-HRP conjugate. The assay sample and buffer are incubated together with GFRBP14-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GFRBP14 concentration since GFRBP14 from samples and GFRBP14-HRP conjugate compete for the anti-GFRBP14 antibody binding site. Since the number of sites is limited, as more sites are occupied by GFRBP14 from the sample, fewer sites are left to bind GFRBP14-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GFRBP14 concentration in each sample is interpolated from this standard curve.

NCBI Description

The product of this gene belongs to a small family of adapter proteins that are known to interact with a number of receptor tyrosine kinases and signaling molecules. This gene encodes a growth factor receptor-binding protein that interacts with insulin receptors and insulin-like growth-factor receptors. This protein likely has an inhibitory effect on receptor tyrosine kinase signaling and, in particular, on insulin receptor signaling. This gene may play a role in signaling pathways that regulate growth and metabolism. Transcript variants have been reported for this gene, but their full-length natures have not been determined to date. [provided by RefSeq, Jul 2008]

Plays a critical role regulating PDPK1 membrane translocation in response to insulin stimulation and serves as an adapter protein to recruit PDPK1 to activated insulin receptor, thus promoting PKB/AKT1 phosphorylation and transduction of the insulin signal. Belongs to the GRB7/10/14 family.
