Intended Use This GPC1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey GPC1. This ELISA kit for research use only!
PRINCIPLE OF THE ASSAY GPC1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GPC1 antibody and an GPC1-HRP conjugate. The assay sample and buffer are incubated together with GPC1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GPC1 concentration since GPC1 from samples and GPC1-HRP conjugate compete for the anti-GPC1 antibody binding site. Since the number of sites is limited, as more sites are occupied by GPC1 from the sample, fewer sites are left to bind GPC1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GPC1 concentration in each sample is interpolated from this standard curve.

Members of the glypican-related integral membrane proteoglycan family (GRIPS) contain a core protein anchored to the cytoplasmic membrane via a glycosyl phosphatidylinositol linkage. GPC1: Cell surface proteoglycan that bears heparan sulfate. Binds, via the heparan sulfate side chains, alpha-4 (V) collagen and participates in Schwann cell myelination. May act as a catalyst in increasing the rate of conversion of prion protein PRPN(C) to PRNP(Sc) via associating (via the heparan sulfate side chains) with both forms of PRPN, targeting them to lipid rafts and facilitating their interaction. Required for proper skeletal muscle differentiation by sequestering FGF2 in lipid rafts preventing its binding to receptors (FGFRs) and inhibiting the FGF-mediated signaling. Associates (via the heparan sulfate side chains) with fibrillar APP-beta amyloid peptides in primitive and classic amyloid plaques and may be involved in the deposition of these senile plaques in the Alzheimer disease (AD) brain. Misprocessing of GPC1 is found in fibroblasts of patients with Niemann-Pick Type C1 disease. This is due to the defective deaminative degradation of heparan sulfate chains.
