Principle of the Assay: GP-130 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GP-130 antibody and an GP-130-HRP conjugate. The assay sample and buffer are incubated together with GP-130-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GP-130 concentration since GP-130 from samples and GP-130-HRP conjugate compete for the anti-GP-130 antibody binding site. Since the number of sites is limited, as more sites are occupied by GP-130 from the sample, fewer sites are left to bind GP-130-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GP-130 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a signal transducer shared by many cytokines, including interleukin 6 (IL6), ciliary neurotrophic factor (CNTF), leukemia inhibitory factor (LIF), and oncostatin M (OSM). This protein functions as a part of the cytokine receptor complex. The activation of this protein is dependent upon the binding of cytokines to their receptors. vIL6, a protein related to IL6 and encoded by the Kaposi sarcoma-associated herpesvirus, can bypass the interleukin 6 receptor (IL6R) and directly activate this protein. Knockout studies in mice suggest that this gene plays a critical role in regulating myocyte apoptosis. Alternatively spliced transcript variants have been described. gp130: gp130 is a ubiquitously expressed type I cytokine family receptor. The receptor systems for IL6, LIF, OSM, CNTF, IL11 AND CT1 utilize gp130 for initiating signal transmission. Binds to IL6/IL6R (alpha chain) complex, resulting in the formation of high-affinity IL6 binding sites, and transduces the signal. Does not directly bind IL6. May have a role in embryonic development. Contains 5 fibronectin type III domains and 1 immunoglobulin-like C2-type domain. Two alternatively spliced isoforms have been described.
