Principle of the assay: gp130 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-gp130 antibody and an gp130-HRP conjugate. The assay sample and buffer are incubated together with gp130-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the gp130 concentration since gp130 from samples and gp130-HRP conjugate compete for the anti-gp130 antibody binding site. Since the number of sites is limited, as more sites are occupied by gp130 from the sample, fewer sites are left to bind gp130-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The gp130 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a signal transducer shared by many cytokines, including interleukin 6 (IL6), ciliary neurotrophic factor (CNTF), leukemia inhibitory factor (LIF), and oncostatin M (OSM). This protein functions as a part of the cytokine receptor complex. The activation of this protein is dependent upon the binding of cytokines to their receptors. vIL6, a protein related to IL6 and encoded by the Kaposi sarcoma-associated herpesvirus, can bypass the interleukin 6 receptor (IL6R) and directly activate this protein. Knockout studies in mice suggest that this gene plays a critical role in regulating myocyte apoptosis. Alternatively spliced transcript variants encoding distinct isoforms have been described. A related pseudogene has been identified on chromosome 17. [provided by RefSeq, Jul 2010]

Binds to IL6/IL6R (alpha chain) complex, resulting in the formation of high-affinity IL6 binding sites, and transduces the signal. Does not directly bind IL6. May have a role in embryonic development. Contains 5 fibronectin type III domains and 1 immunoglobulin-like C2-type domain. Two alternatively spliced isoforms have been described.
