Principle of the Assay: GDF-11 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GDF-11 antibody and an GDF-11-HRP conjugate. The assay sample and buffer are incubated together with GDF-11-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GDF-11 concentration since GDF-11 from samples and GDF-11-HRP conjugate compete for the anti-GDF-11 antibody binding site. Since the number of sites is limited, as more sites are occupied by GDF-11 from the sample, fewer sites are left to bind GDF-11-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GDF-11 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the bone morphogenetic protein (BMP) family and the TGF-beta superfamily. This group of proteins is characterized by a polybasic proteolytic processing site which is cleaved to produce a mature protein containing seven conserved cysteine residues. The members of this family are regulators of cell growth and differentiation in both embryonic and adult tissues. Studies in mice and Xenopus suggest that this protein is involved in mesodermal formation and neurogenesis during embryonic development. [provided by RefSeq, Jul 2008]
