Principle of the Assay: FPN ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FPN antibody and an FPN-HRP conjugate. The assay sample and buffer are incubated together with FPN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FPN concentration since FPN from samples and FPN-HRP conjugate compete for the anti-FPN antibody binding site. Since the number of sites is limited, as more sites are occupied by FPN from the sample, fewer sites are left to bind FPN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FPN concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a cell membrane protein that may be involved in iron export from duodenal epithelial cells. Defects in this gene are a cause of hemochromatosis type 4 (HFE4). [provided by RefSeq, Jul 2008]
Uniprot Description
SLC40A1: May be involved in iron export from duodenal epithelial cell and also in transfer of iron between maternal and fetal circulation. Mediates iron efflux in the presence of a ferroxidase (hephaestin and/or ceruloplasmin). Defects in SLC40A1 are the cause of hemochromatosis type 4 (HFE4). HFE4 is an autosomal dominant iron-loading disorder characterized by early iron accumulation in reticuloendothelial cells and a marked increase in serum ferritin before elevation of the transferrin saturation. Belongs to the ferroportin (FP) (TC 2.A.100) family. SLC40A subfamily.

Product Notes
The Porcine FPN slc40a1 (Catalog #AAA749658) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA749658 ELISA Kit recognizes Porcine FPN. It is sometimes possible for the material contained within the vial of "Ferroportin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
