Description: Recombinant human FLT3 (Y591-V592insVDFREYEYD) or FLT3-ITD ( (571-993) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag.
Scientific Background: Fms-related tyrosine kinase 3 (FLT3) is a gene that encodes for a tyrosine kinase that activates pathways in hematopoietic cells important in cellular proliferation. FLT3 is frequently mutated in acute myeloid leukemia (AML), myelodysplastic syndromes, other hematologic malignancies, and colorectal cancer. FLT3 mutations are observed in 24.3% of AML, of which internal tandem duplications (ITD) are the most frequent. FLT3 ITD occurs when sequences of less than ten to several hundred bases in length are repeated within the juxtamembrane domain are always œin-frame.

NCBI Description

This gene encodes a class III receptor tyrosine kinase that regulates hematopoiesis. This receptor is activated by binding of the fms-related tyrosine kinase 3 ligand to the extracellular domain, which induces homodimer formation in the plasma membrane leading to autophosphorylation of the receptor. The activated receptor kinase subsequently phosphorylates and activates multiple cytoplasmic effector molecules in pathways involved in apoptosis, proliferation, and differentiation of hematopoietic cells in bone marrow. Mutations that result in the constitutive activation of this receptor result in acute myeloid leukemia and acute lymphoblastic leukemia. [provided by RefSeq, Jan 2015]

FLT3 -/- mice have an impaired developmental capacity of primitive hematopoietic progenitor cells of all lineages with the greatest impact on lymphopoietic precursors. Activating mutations found in one third of cases of acute myeloid leukemia (AML), as well as in acute lymphoblastic leukemia, acute promyelocytic leukemia and myelodysplastic syndrome.
