Principle of the assay: FCAR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FCAR antibody and an FCAR-HRP conjugate. The assay sample and buffer are incubated together with FCAR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FCAR concentration since FCAR from samples and FCAR-HRP conjugate compete for the anti-FCAR antibody binding site. Since the number of sites is limited, as more sites are occupied by FCAR from the sample, fewer sites are left to bind FCAR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FCAR concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the immunoglobulin gene superfamily and encodes a receptor for the Fc region of IgA. The receptor is a transmembrane glycoprotein present on the surface of myeloid lineage cells such as neutrophils, monocytes, macrophages, and eosinophils, where it mediates immunologic responses to pathogens. It interacts with IgA-opsonized targets and triggers several immunologic defense processes, including phagocytosis, antibody-dependent cell-mediated cytotoxicity, and stimulation of the release of inflammatory mediators. Multiple alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Jul 2008]
