Principle of the Assay: This kit is based on sandwich enzyme-linked immune-sorbent assay technology. Anti-IL-1R1 polyclonal antibody is precoated onto 96-well plates and the biotin conjugated anti-IL-1R1 polyclonal antibody is used as the detection antibody. The standards, test samples, and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with wash buffer. TMB substrates are used to visualize the HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow is proportional to the IL- 1R1 amount of sample captured in plate. The O.D. absorbance is read at 450nm using a microplate reader, and then the concentration of IL-1R1 can be calculated.
Background: Interleukin 1 receptor, type I (IL1R1) also known as CD121a (Cluster of Differentiation 121a), is an interleukin receptor. IL1R1 also denotes its human gene. Helicobacter pylori (Hp) is an environmental inducer of gastritis and gastric cancer (GC). The immune response to Hp and the associated changes in somatic gene expression are key determinants governing the transition from gastritis to GC. Disruption of the HNF4alpha/IL-1R1/IL-1beta/NF-kappaB circuit during Hp infection maybe an effective means of preventing the associated GC. Signaling through the IL-1-receptor type 1 (IL-1R1), IL-1 is required for initiation and maintenance of diverse activities of the immune system. A second receptor, IL-1R2, blocks IL-1 signal transduction. The functions of the IL-1R2 as a "decoy-receptor" sequestrating paracrine IL-1 extracellularly and intracellularly by engaging IL-1IRAcP, thus depriving IL1-R1 molecules of their extracellular and intracellular ligands. Expression of IL1-R2 by HRS cells seems to contribute to local and systemic modulation of immune function in HL.
