Intended Uses: This HTR2A ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse HTR2A. This ELISA kit for research use only!

Principle of the Assay: HTR2A ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-HTR2A antibody and an HTR2A-HRP conjugate. The assay sample and buffer are incubated together with HTR2A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HTR2A concentration since HTR2A from samples and HTR2A-HRP conjugate compete for the anti-HTR2A antibody binding site. Since the number of sites is limited, as more sites are occupied by HTR2A from the sample, fewer sites are left to bind HTR2A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HTR2A concentration in each sample is interpolated from this standard curve.

APOA1: Participates in the reverse transport of cholesterol from tissues to the liver for excretion by promoting cholesterol efflux from tissues and by acting as a cofactor for the lecithin cholesterol acyltransferase (LCAT). As part of the SPAP complex, activates spermatozoa motility. Interacts with APOA1BP and CLU. Component of a sperm activating protein complex (SPAP), consisting of APOA1, an immunoglobulin heavy chain, an immunoglobulin light chain and albumin. Interacts with NDRG1. Major protein of plasma HDL, also found in chylomicrons. Synthesized in the liver and small intestine. The oxidized form at Met-110 and Met-136 is increased in individuals with increased risk for coronary artery disease, such as in carrier of the eNOSa/b genotype and exposure to cigarette smoking. It is also present in increased levels in aortic lesions relative to native ApoA-I and increased levels are seen with increasing severity of disease. Belongs to the apolipoprotein A1/A4/E family.
